Ribosomes, Peptides and Antibiotic Resistance

چکیده
A new approach for studying functional rRNA fragments has been developed based on using a plasmid library expressing random fragments of rRNA, A 34 nucleotide long fragment of Escherichia coli 23S rRNA has been identified that renders cells resistant to erythromycin, when expressed in vivo. The rRNA fragment contains a five codon long open reading frame, initiating at GUG and terminating at UAA, with a Shine-Dalgamo sequence located at an appropriate distance from the initiator codon. Translation of this mini-gene is required for the observed erythromycin resistance. Experiments with in vitro translated, or synthetic, peptide indicate the ribosome as a likely target for the action of the identified rRNA-encoded peptide, which apparently remains associated with the ribosome after completion of its translation. The known properties of the rRNA-encoded peptide are compared with information about other functionally active short peptides that can be involved in regulation of translation.

Antibiotic resistance peptides: interaction of peptides conferring macrolide and ketolide resistance with Staphylococcus aureus ribosomes: conformation of bound peptides as determined by transferred NOE experiments.
Two antibiotic resistance peptides, the E-peptide (MRLFV) and the K-peptide (MRFFV) conferring macrolide and ketolide resistance, respectively, were studied in the complex state with bacterial Staphylococcus aureus ribosomes. Interactions of antibiotic resistance peptides with ribosomes were investigated using two-dimensional transferred nuclear Overhauser effect spectroscopy (TRNOESY), suggest...
متن کاملPeptide-mediated macrolide resistance reveals possible specific interactions in the nascent peptide exit tunnel.
Expression of specific short peptides can render cells resistant to macrolide antibiotics. Peptides conferring resistance to structurally different macrolides including oleandomycin, azithromycin, azaerythromycin, josamycin and a ketolide cethromycin were selected from a random pentapeptide expression library. Analysis of the entire collection of the resistance peptides allowed their classifica...
متن کاملMacrolide antibiotics allosterically predispose the ribosome for translation arrest.
Translation arrest directed by nascent peptides and small cofactors controls expression of important bacterial and eukaryotic genes, including antibiotic resistance genes, activated by binding of macrolide drugs to the ribosome. Previous studies suggested that specific interactions between the nascent peptide and the antibiotic in the ribosomal exit tunnel play a central role in triggering ribo...
متن کاملRibosomal alterations contribute to bacterial resistance against the dipeptide antibiotic TAN 1057.
TAN 1057-resistant Staphylococcus aureus and Escherichia coli strains were selected to elucidate the mechanism of resistance and the mode of action of this dipeptide antibiotic. Cell-free translation with isolated ribosomes and S150 fractions from sensitive and resistant S. aureus strains demonstrated that alterations in the ribosomes contribute to the resistance of the bacteria.
متن کاملRegulation of antibiotic-resistance by non-coding RNAs in bacteria.
Antibiotic resistance genes are commonly regulated by sophisticated mechanisms that activate gene expression in response to antibiotic exposure. Growing evidence suggest that cis-acting non-coding RNAs play a major role in regulating the expression of many resistance genes, specifically those which counteract the effects of translation-inhibiting antibiotics. These ncRNAs reside in the 5'UTR of...
متن کاملLocalization of five antibiotic resistances at the subunit level in chloroplast ribosomes of Chlamydomonas.
The chloroplast ribosomes from five antibiotic resistant strains of Chlamydomonas, each carrying one mutant gene mapping in chloroplast DNA, have been shown to be resistant to the corresponding antibiotic in a poly(U)-directed amino-acid incorporating assay system. The alteration conferring resistance was localized to the 30S subunit in ribosomes from streptomycin, neamine, and spectinomycin re...
